  • Background:

    The mitochondrial AOX (alternative oxidase) of the unicellular green alga Chlamydomonas reinhardtii is encoded by two different genes, the Aox1 and Aox2. The  alternative respiratory pathway is comprised of a single homodimeric protein – AOX – and functions as a mechanism to decrease the formation of reactive oxygen species (ROS) produced during respiratory electron transport.  Alternative oxidase expression is  influenced by different stress stimuli.

    Used on chlamydomonas. Good signal.

    Loaded 20 ug of C. reinhardtii whole cell extract from freeze thaw method. Blocked with TBS+0.1%Tween, 5% milk for 1 hr at rm temp or overnight at 4 deg C. Antibody diluted 1:10,000 in TBS+0.1%Tween, 2% milk. Primary incubation overnight with gentle agitation at 4 deg C. 5 washes in between primary and secondary, volume for all antibody incubations and washes was 25 ml, washes at room temp. Immunoblot with HRP-conjugated secondary (1:20,000, 0.1%TBST) was clean with these conditions, however, longer exposure will yield a few additional bands, one at ~60 kDa and another at ~30 kDa (12% polyacrylamide).
